Does the rear prop shaft turn when trying to drive? if yes, then the fault lies with the back axle (diff, half shafts, drive flanges). If no, then it's most likely the transfer box.
Thinking about it a bit more, the fact that the handbrake works suggests that the fault is in the transfer box, too. I thought that but then if the handbreak locks the back axel it proves nothing as the back wheels will be locked if it's the transfer box and the transfer box if it's the rear diff, further investigation has suggested the diff lock isn't engaging, link looks good so changing the transmission fluid to see if any metal appears

You drive (without diff lock) your Defender and get a vibration. Then you take the front prop off to check it. But now have no drive to the rear, and non operating diff lock.
This can’t happen. If you have no drive to rear, and non operating centre diff lock you wouldn’t be able to drive the vehicle. |

Yes but I didn't know the diff lock wasn't engaging, addressing this problem now

The diff lock switch is acting on the locking hub, so assuming that the wiring is good and dash light is also good, the light would come on when it is truly locked.
The diff lock lever is right above the front output flange. Easiest access is through the middle hatch in the seatbox, under the cubby box or middle seat. I am assuming you remove the front prop shaft to isolate the vibration issue. Correct? Propshafts have balancing plates welded to them, and if they fall off, they can vibrate. Prop shafts both have plates both and are sound, still can't get the diff light on, I've checked I have drive to both shafts and I do, sitting still both torq up when power applied, when the rain stops, tomorrow, I shall remove the switch and test it after that I am lost for a solution to the diff lock issue and none the wiser on my vibration issue

You’ve removed the front prop, and now the trans box is spinning the easiest route, the front drive flange, as you’ve removed the prop. So no diff or axle problems, just need to sort the diff lock. Take the covers off from inside the defender and expose the top of the trans case and you will see the operation rod. Easy to remove cover and check that grub screw that operates inside shaft is tight and moving shaft. |

^^^ IIRC grub screw is only on the high/low shaft.

Just wind out the diff lock switch a few threads, it's stopping the lock engaging. Cheers
