With the front doors, the locking button is directly above the exterior handle and the key barrel. With the second row, it’s on the other end of the door.
I have to say, while I’ve had my doors apart many times over the years, I’m not familiar enough with the mechanism to know if this would work, even if you did have a barrel in the handle? Is this a known option? I've just had a good look at the parts diagrams and I don't think this will work. The rear door latches lock with a central locking actuator and/or with the button. The front door latch has an extra linkage that goes to the lock barrel in the handle. Even if you fitted a lock barrel to the existing handle, or a front handle mounted on the rear door, I think you'd have a real challenge on your hands getting all the fiddly little rods to line up and do their job. I wouldn't say it is impossible but I'd question the time & hassle factor versus benefit here.
